Where to Get the Silksong Flexile Spines Quest?

You can get the Flexile Spines quest from an NPC named Seamstress, whom you will find in the area marked in white in the lower right parts of Far Fields. When you interact with her, she will ask if you want to accept the quest. Accept it and prepare to set off.

Fortunately, we don't have to travel far to do this quest, as the creatures needed to complete it are located in the area we've marked in green on the map.
To complete the Flexile Spines quest, you need to kill the creatures named Hokers, shown in the image above, and collect the spines they drop when they attack you one by one.
After collecting 25 of these spines, go to Seamstress and report that the quest is finished, and she will present you with your reward.

When you return to Seamstress after collecting everything you need, you will receive a power-up called Drifter’s Cloak as a reward. This power-up allows you to ascend through air currents and use your cloak like a parachute when falling from high places.
The Drifter’s Cloak is especially necessary to get past some of the platforming sections in the later parts of Silksong. For this reason, we recommend that you do not skip this quest and complete it at the first opportunity.
